Collection Info

  One nasopharyngeal (not throat, nose, or nasal) specimen collected 
on a rayon swab with an aluminum or plastic shaft should be placed 
in transport medium such as Stuart's with charcoal or Amies with 
charcoal (Transwab Nasopharyngeal with Charcoal System) for shipping. 

Note:     1.  The following specimens may be inhibitory to PCR testing 
                       and will be rejected:
                       A.  Calcium alginate swabs
                       B.  Cotton-tipped swabs
                       C.  Swabs sent in gel transport medium
                       D. Swabs sent in viral or universal transport medium

                  2.  Stuart's or Amies medium without charcoal will also be 

                       rejected. 

Alternatively, a nasopharyngeal (not throat) aspirate/wash or nasal 
aspirate/wash can be collected and transported in a screw-capped, 
sterile container within 24 hours of collection. Send specimen refrigerated.

Specimen Acceptability

Methods
Bacterial nucleic acid is extracted (IsoQuick Nucleic Extraction) from nasopharyngeal specimens. Rapid PCR amplifies target sequences of the IS481 and IS1001 genes of Bordetella pertusiss and Bordetella parapertusiss respectively using the LightCycler(TM) System (Roche Molecular Biochemicals). Fluorescence Resonance Energy Transfer (FRET) technology allows the real-time detection of specific PCR products.
